| Resumo: | In order to record neuronal activity, neuroscientists can rely on a wide range of techniques or modalities. Each modality comes with its own advantages and disadvantages, specifically in terms of resolution (temporal and spatial) and invasiveness. In this seminar, I will first introduce the different modalities that we can use to record from neurons, specifically focusing on the patch-clamp technique, extracellular electrodes, and fluorescent indicators. After covering the pros and cons of each modality, we will see how a multimodal approach allows us to expand the potential of single modalities to unravel mechanisms at the sub-cellular, cellular, and network scales. First, we¿ll see how we can combine extracellular electrophysiology and calcium imaging to identify monosynaptic connections and characterize responses of single dendritic spines. Next, I will present the benefits of combining the patch-clamp technique with extracellular recordings, both in order to better understand and model single neurons and to investigate network dynamics. |
